Most of these trophies will naturally be achieved during your game. However, you'll need to complete one playthrough of the game on any difficulty first to unlock Professional Mode as well as the other Resident Evil 4 Separate Ways unlocks.
Ultimately, this means that you'll need to have completed the DLC at least twice to get all of the trophies/achievements.
