The Whistle emote can be earned on the Lost Ark Azure Wind Island, which is north of Yorn, and south of Rohendel. It's also near the Lost Ark Phantom Wing Island, which is directly north-east. Furthermore, it's recommended you reach equipped item level 802 before venturing here, which is quite far into the end-game.

The Lost Ark Whistle emote is a reward for gaining level three reputation in the Ruffians in the Ground quest, which is part of Una's Daily Tasks that unlocks at combat level 50. Pressing ALT+J will allow you to see the full Una Tasks list, along with your progress in any of the tasks. 

 

Ruffians in the Ground tasks you with culling the island of boars, and you need to repeat it multiple times until you gain level three recognition and earn the emote, along with a stat increase potion and giant's heart. You also need to start the Azure Wind Island questline and reach The Main of the Azure Wind before taking part in this task.

Once you have the emote, just activate it from your inventory, and then you perform it by going into the emote menu or typing /whistle in the chat.
